Genetic diversity analysis of Chinese Leishmania isolates and development of L. donovani complex-specific markers by RAPD

Abstract Background Leishmaniasis is one of the most neglected tropical diseases in world and remains endemic some underdeveloped regions, including western China. The phylogeny classification Chinese Leishmania has not been completely clarified to date, especially within ( L. ) donovani complex, although phylogenetic analyses based on a series gene markers have performed. More analytic methods data are still needed. Random amplified polymorphic DNA (RAPD) technology can sensitively identify slight intraspecific differences, it powerful tool seek species-specific markers. This work attempted isolates from diverse geographic regions at genomic level. Meanwhile, specific complex were also developed by RAPD. Three SCAR i.e., 1-AD17, 2-A816 3-O13, successfully obtained study. Through bioinformatic analyses, Marker 1-AD17 may more specificity for PCR detection VL, 3-O13 potential encode protein. Conclusions results verified undescribed species causing visceral leishmaniasis (VL) China unique clade distinguished revealed there differentiation among . identification -specific help provide foundation future research attempting develop new diagnostic VL functions.
